The InviMag Stool DNA Mini Kit/ KFmL allows for rapid and economical purification of genomic and microbial DNA from up to 200 mg of stool sample or about 1 g sample in 8 ml Stool Stabilizer using magnetic beads. The InviMag Stool DNA Mini Kit/ KFmL has been designed for semi-automated preparation on the KingFisher mL workstation. The kit allows the isolation of host DNA from colon epithelial cells, parasite DNA, bacterial DNA or DNA from gastrointestinal pathogens. Furthermore, it is possible to extract nucleic acids from food and feed residues of plant or animal origin from the stool sample.

Overview

A rigorous pre-lysis step with optimized pre-lysis buffer under high temperatures is followed by a pre-incubation of the sample with InviAdsorb to remove PCR inhibitors outside of the robotic platform. Undissolved particles and PCR inhibitors bound to InviAdsorb are removed by a centrifugation step. Besides the pre-filling of all cavities, all following processing steps are performed by the robot, like Proteinase K digestion and the binding of DNA to magnetic particles, followed by washing steps and the final elution. The choice of different lysis conditions allow for the enrichment or a reduction of the content of bacterial DNA in the total DNA in favor of human DNA.
The purified high quality DNA is ready to use for subsequent downstream applications or can be stored at –20 °C for subsequent use:

In addition to the DNA isolation method, STRATEC Molecular offers an innovative amplification enhancer for DNA from fecal samples. The use of the I-Solution enables the improvement of suboptimal PCR reactions.
